Stilbenoid compounds inhibit NF-κB-mediated inflammatory responses in the Drosophila intestine

چکیده

Introduction Stilbenoid compounds have been described to anti-inflammatory properties in animal models vivo , and shown inhibit Ca2+-influx through the transient receptor potential ankyrin 1 (TrpA1). Methods To study how stilbenoid affect inflammatory signaling we utilized fruit fly, Drosophila melanogaster as a model system. induce intestinal inflammation fed flies with irritant dextran sodium sulphate (DSS). Results We found that DSS induces severe changes bacteriome of intestine, this dysbiosis causes activation NF-κB transcription factor Relish. taken advantage DSS-model pinosylvin (PS) monomethyl ether (PSMME). With help approaches, identified PS PSMME be (TrpA1)-dependent antagonists NF-κB-mediated immune responses . also computationally predicted putative antagonist binding sites these at TrpA1. Discussion Taken together, show stilbenoids intestine can used alleviate chemically induced

siRNA-mediated silencing of Wnt5a regulates inflammatory responses in atherosclerosis through the MAPK/NF-κB pathways.

Previous studies have demonstrated that the aberrant expression of Wnt5a occurs in atherosclerotic lesions. However, the precise role of Wnt5a in the pathogenesis of atherosclerosis remains largely unknown. The present study was undertaken to determine whether the RNA interference of Wnt5a in vivo by adenovirus (Ad)-mediated small interfering RNA (siRNA) transfection is capable of inhibiting th...

hCLOCK Causes Rho-Kinase-Mediated Endothelial Dysfunction and NF-κB-Mediated Inflammatory Responses

BACKGROUND The human Circadian Locomotor Output Cycle protein Kaput (CLOCK) gene was originally discovered as a regulator of essential human daily rhythms. This seemingly innocuous gene was then found to be associated with a multitude of human malignancies, via several biochemical pathways. We aimed to further investigate the role of hCLOCK in the hypoxia-oxidative stress response system at the...

Regulation of the NF-κB-Mediated Transcription of Inflammatory Genes

The NF-κB family of transcription factors plays a central role in the inducible expression of inflammatory genes during the immune response, and the proper regulation of these genes is a critical factor in the maintenance of immune homeostasis. The chromatin environment at stimulus-responsive NF-κB sites is a major determinant in transcription factor binding, and dynamic alteration of the chrom...

Overexpression of Sirtuin 6 suppresses cellular senescence and NF-κB mediated inflammatory responses in osteoarthritis development

The aim of our study was to evaluate if Sirt6, a NAD + dependent histone deacetylase, plays a protective role in cartilage degeneration by suppressing cellular senescence and inflammatory responses. The expression level of sirt6 in normal and OA human knee articular cartilage was compared by immunofluorescence and western blotting. The effect of sirt6 overexpression on replicative senescence of...

Toll-like receptor 2–mediated NF-κB inflammatory responses in dry eye associated with cGVHD

PURPOSE Dry eye syndrome is one of the most common pathological manifestations in patients with chronic graft-versus-host disease (cGVHD). When dry eye occurs in association with cGVHD, the local inflammation is secondary to the systemic onset of inflammation. Toll-like receptor 2 (TLR2) signaling is thought to be essential for the inflammatory response and for immune disorders. The aim of this...
